Learn More About Hyperpolarization

Hyperpolarization techniques overcome the inherent sensitivity limitations of magnetic resonance by dramatically boosting nuclear spin polarization well beyond thermal equilibrium levels. Two main approaches are widely used: Dynamic Nuclear Polarization (DNP), which transfers polarization from unpaired electrons to nuclei under cryogenic conditions before a rapid dissolution step, and Parahydrogen Induced Polarization (PHIP), which transfers polarization directly from parahydrogen, yielding a polarized sample in seconds to minutes without a radical doping agent.

In dissolution DNP, the sample is kept cold (typically below 4 K) under a strong magnetic field while irradiated by microwaves, then rapidly dissolved in a hot, pressurized deuterated solvent and transferred for immediate NMR or MRI acquisition. The pyruvate- and urea-based substrates listed above are among the most widely used hyperpolarized probes for in vivo 13C metabolic imaging, providing insight into substrate localization, biochemical transformations, and intracellular pH.
